The exact role of mitochondria in the pathogenesis of Alzheimer’s disease (AD) is not well understood. Gene dependency is a common occurrence in biological processes, where many transcription factors that regulate their targets rely on other regulators. Therefore, identifying gene dependencies after observing phenotypic changes can help us better understand the regulatory mechanisms behind such phenotypes. To address this issue, several methods have been proposed to construct Gene Regulatory Networks (GRNs) based on gene expression data. However, these GRNs can only represent static regulatory relationships and cannot identify gene dependencies that are essential for investigating biological mechanisms behind a specific phenotype or disease. In our previous work, we presented a method to construct regulatory networks that reveal gene dependence during phenotypic changes, which has proven successful in breast cancer precision medicine. In this study, we used transcriptomic data from AD patients and controls to construct gene dependency networks for exploring mitochondrial-related mechanisms in AD.
